Intervenolin (ITV) produced by Nocardia sp. ML96-86F2 is a unique natural compound having both anti-Helicobacter pylori activity and anti-tumor activity. We are developing potent drug candidates through AI-based chemical syntheses of ITV analogues.

(1) Anti-H. pylori drug candidates
More than 50% of world population is infected with H. pylori, a cause of gastric cancer. The triple therapy containing two antibiotics is currently used to eradicate the H. pylori infection, but its drug-resistant strains and its adverse effect against intestinal biota are serious problems. ITV analogues are very effective against such drug-resistant H. pylori strains and show selective anti-H. pylori activity without effect on intestinal biota through inhibition of DHODH, a critical enzyme of pyrimidine synthesis. ITV analogues eradicate H. pylori infection in animal models.

(2) Anti-tumor drug candidates
Cancer cells exist with surrounding stroma containing such as fibroblasts. The stroma can promote cancer cell growth and metastasis, suppress tumor immunity, and resist against anti-tumor drugs. ITV analogues are designed to affect the interaction between cancer cells and stroma. ITV analogues inhibit mitochondrial complex I and exert anti-tumor activity through acidification of intracellular and extracellular environment of cancer cells.
